dc.descriptionThe vascular wilt of Babaco (VWB), caused by Fusarium oxysporum is a sickness of great economic importance because of the great losses it causes to the Babaco crops. This thesis has allowed for the study of molecular and taxonomic characterization of the causal agents of the vascular wilt of Babaco Vasconcellea heilbornii Badillo. var. pentagonal, in Loja and Saraguro county. Samples of the root of Babaco with symptoms of VWB were gathered during the months of January and February 2013. Later the 11 fungal isolates were characterized morphologically and culturally considering the following parameters: type of mycelium, color of the colony, radial growth measurement, quantification and measurement of the reproductive structures (macro and micro conidia) and counting of the conidia. Then molecular techniques were applied for identification at the genus and species level. In the same way, the 13 bacterial isolates were characterized morphologically, culturally and biochemically (Growth, Color, Height, Border, Presence of Mucus, Gram Staining, Growth in King B Environment, Catalase Test, Growth in McConkey and Kliger environments , and fluorescence presence). The results obtained show 11 fungal isolates associated with the VWB which have morpho-cultural characteristics indicative of the Fusarium sp. genus. A high variability was observed among the isolates in the different places where they were gathered, with respect to the pigmentation of the colonies, type of mycelium and the measurements of the reproductive structures. According the radial growth, three categories of classification were identified: three isolates with slow growth, five with the medium growth, and three with rapid growth. The genetic identification through the sequencing ITS obtained with the universal primers and their later comparison with the sequences previously reported and identified in the NCBI, allowed four groups to be identified: Group 1; Fusarium sp LF236. Group 2; Fusarium oxysporum strain 2T12JO1A. Group 3; Fusarium oxysporum isolate IBSD-F.O. Group 4; Fusarium oxysporum f. sp. ciceris, establishing more than one species of Fusarium associated with the VWB. The morpho-cultural and biochemistry parameters did not detect any variability among the 13 bacterial isolates associated with the VWB. These parameters allowed the negative-Gram bacteria to be identified possibly of the Pseudomonas genus. The results obtained through the molecular identification and the alignment of sequences, discarded the presence of the species of the Pseudomonas genus in the different bacterial isolates. The data indicate the presence of bacteria that correspond to the Enterobacteriaceae family: Enterobacter amnigenus, Lelliottia amnigena, Klebsiella pneumoniae, Kluyvera intermedia, Klebsiella sp., Enterobacter sp., Enterobacteriaceae bacterium. These bacteria cause infection in human beings but are not yet reported as the casual agents of phytosanitary diseases in the Babaco. However, diverse investigations enunciate that the bacteria of the Enterobacter genus as promoters of vegetable growth and casual agents of phytosanitary sickness in Onions Allium cepa and in mulberry Morus alba.es_ES
dc.description.abstractLa Marchitez Vascular del Babaco (MVB), causada por Fusarium oxysporum es una enfermedad de gran importancia económica debido a las severas pérdidas que produce en el cultivo de Babaco. Esta tesis permitió estudiar la caracterización taxonómica y molecular de los agentes causales de la marchitez vascular del Babaco Vasconcellea heilbornii Badillo. var. pentágona, en los cantones de Loja y Saraguro. Se recolectaron muestras de raíz de Babaco con síntomas de MVB durante los meses de enero y febrero de 2013. Posteriormente los 11 aislados fúngicos se caracterizaron morfológica y culturalmente teniendo en cuenta los siguientes parámetros: tipo de micelio, color de la colonia, medición del crecimiento radial, cuantificación y medición de las estructuras reproductivas (macro y microconidias) y el conteo de conidios, seguidamente se aplicaron técnicas moleculares para la identificación a nivel de género y especie. Asimismo los 13 aislados bacterianos se caracterizaron morfológica, cultural y bioquímicamente (Crecimiento, Color, Elevación, Borde, Presencia de mucus, Tinción Gram, Crecimiento en medio King B, Prueba de la catalasa, crecimiento en medio McConkey, crecimiento en medio Kliger y presencia de fluorescencia). Los resultados obtenidos muestran 11 aislados fúngicos asociados a la MVB, que poseen características morfo-culturales indicativas al género Fusarium sp. Se observó una alta variabilidad entre los aislados en diferentes sitios de colecta, respecto a la pigmentación de las colonias, tipo de micelio y en la medición de las estructuras reproductivas. Según el crecimiento radial se identificaron tres categorías de clasificación: tres aislados con crecimiento lento, cinco de crecimiento medio y tres con crecimiento rápido. La identificación genética mediante la secuenciación ITS obtenidas con primers universales y su posterior comparación con secuencias previamente reportadas e identificadas en la NCBI, permitió identificar cuatro grupos: Grupo 1; Fusarium sp LF236. Grupo 2; Fusarium oxysporum strain 2T12JO1A. Grupo 3; Fusarium oxysporum isolate IBSD-F.O. Grupo 4; Fusarium oxysporum f. sp. ciceris, estableciendo más de una especie de Fusarium asociada a la MVB. Los parámetros morfo-culturales y bioquímicos no detectaron variabilidad alguna en los 13 aislados bacterianos asociados a la MVB, estos parámetros permitieron identificar bacterias Gram-negativas posiblemente del género Pseudomonas. Los resultados obtenidos mediante la identificación molecular y el alineamiento de secuencias, descartaron la presencia de las especies del género Pseudomonas en los diferentes aislados bacterianos, los datos indican la presencia de bacterias que corresponden a la familia Enterobacteriaceae: Enterobacter amnigenus, Lelliottia amnigena, Klebsiella pneumoniae, Kluyvera intermedia, Klebsiella sp., Enterobacter sp., Enterobacteriaceae bacterium. Estas bacterias son causantes de infecciones en los seres humanos, y aún no son reportadas como causantes de enfermedades fitosanitarias en Babaco, sin embargo diversas investigaciones enuncian a bacterias del género Enterobacter como promotoras del crecimiento vegetal y causantes de enfermedades fitosanitarias en cebolla Allium cepa y en morera Morus alba.es_ES
